mRNA Display-based Self-Assembling Protein Microarray Development

The new generation of self-assembly protein chip can express and fix proteins in real time and in situ on the chip by using cell-free expression system and DNA fixation technology, effectively solving the problems of preparation and preservation of traditional protein chips, and has been initially used in large-scale protein-protein interaction screening and identification of immune dominant antigens. Introduction of Protein Microarray

Two basic features are required for function-based protein molecular screening: first, functional information (natural structure of protein) is obtained through high-throughput analysis of rich protein libraries; Secondly, the genetic information (DNA sequence) is decoded from the analyzed proteins by linking the expressed phenotype with its coding genotype. Protein microarray provides a means for high-throughput analysis and molecular display technology, in which genotype and phenotype are physically linked, providing sequence information for identification purposes. However, in order to be truly effective, future complex proteomics research will need to carry out molecular screening for very large populations (super protein libraries) in a side-by-side manner and under the conditions of required functions.

The integration of molecular display technology, such as mRNA display, is known as the most simplified molecular display and a powerful means of screening the most diverse mutant library.
