Overview of Immune Antibody Library

The immune antibody library is constructed from B cells of immune or infected individuals targeting specific antigens of interest. After exposure to antigen, in vivo affinity maturation and V gene hypermutation contribute to the formation of a specific gene pool for antigen. Therefore, antibodies isolated from immune libraries usually exhibit higher affinity in the nanomolar range. Like naïve antibody libraries, antibody genes can also be isolated from PBMC, bone marrow, lymph nodes or spleen. IgG is usually used to build immune antibody libraries, while IgA and IgE are sometimes used for libraries based on mucosa, parasites and allergies. Immune antibody libraries are usually customized for a specific set of antigens associated with a specific infection. Although the separation of binders for closely related antigens has been reported, the immune library is usually not used for different antigens, because it is generally believed that the skewed gene library will mean the key application of the immune library.
